You've been told multiple times now that the aspect ratios need to match in order to even be semi-accurate. In my last post in this thread I said you divide the width by the height to find the aspect ratio, so let me do that for you now since you refuse to do it yourself.
3840/2160 = 1.78
800/600 = 1.33
Do those numbers match?

In order to run different resolutions, again, the aspect ratios need to match. You will also need to adjust your Repeater Profile to scale from the center.

Because you're trying to run the game clients at such a high resolution and Diablo III takes a lot of system resources. A lot of people think they have super computers until they begin multiboxing multiple game clients. Once they do they quickly learn that their computers aren't as 'super' as they thought. I doubt 4K is going to look as good as you think in Windowed mode -- I'm sure you've been using it in Fullscreen mode and it's nice, but downsampling doesn't work all that great when windowed. Either way, there's no easy way to do this because you want something so super custom and you sound like you're incredibly new to the program and how Window Layouts work, but even then I don't know exactly how to go about creating the layout you want. You can try this if you'd like:
1) Turn off Instant Swapping in your Window Layout
2) Create a fifth region and set its size to 3840x2160
3) On the Swap Groups tab set the fifth region to be the Reset Region
4) Adjust regions 2, 3, and 4 to have a 1.78 (16:9) aspect ratio
5) Scale your Repeater Profile from the center
6) Export and launch your Character Set
Also, Diablo III is a game that doesn't like its resolution to be messed with and you might not be able to choose 4K @ 60 FPS -- You may have to end up running it at 30 FPS. You may also have to toggle between display modes in order to get 4K to even work, and I have no idea how well it's going to work if you ever expect to swap windows around.

You want to be using Windowed Fullscreen - viewtopic.php?f=40&t=3206
