Issue with FTL Assist

Quite stumped.

I have created a new character set for boosting.
This is called "Boost Druids".
Everyone will assist (FTL Assist Me) my Slot 3 and Slot 4 character.
No one will assist the Slot 1, Slot 2 and Slot 5 characters.

They use the same FTL Assist Me mapped key that works fine for all my other character sets.
I created another assist called Assist Me, because in the past I've had mapped keys that fail, but when deleted and made with identical syntax they then work.
In this case, the new mapped key has exactly the same behavior.

Everyone has a unique FTL key combination.
My FTL Assist Me sends "F" to the game.
This is not bound to anything in the game on any character on the team.

OK, your "FTL Assist Me" Mapped Key and your "Assist Me" Mapped Key (both in the General Key Map) have the same behaviour because the Assist Me Mapped Key just calls the FTL Assist Me Mapped Key with a Do Mapped Key Action. That is expected based upon how you have things set up.

The fact that when using FTL Assist Me everyone will assist Slots 3 and 4 but no-one will Assist Slots 1, 2 and 5 is strongly suggestive of conflicting keybinds in WoW. There are two possibilities for fixing this if that is indeed the issue:

1. Change the Key Combination used in the WoW Macro Action in FTL Assist Me from F to something else. I'd suggest Backspace.
2. Use the method described here to ensure that the following are not bound in WoW: Shift+F, RightShift+F, Ctrl+Shift+F.

Changing the key that is sent to Warcraft, from F to Backspace, did fix this issue.
Weird though, I didn't have any wow binds on F, Shift F, Ctrl F, Alt F or similar (right modifiers).

It could potentially have been a keybind in another addon conflicting but then I would expect a warning from ISBoxer in the chat frame when WoW's UI was loaded.
