It is probably not significantly beneficial to assign 4 cores to the 1 character while restricting 4 characters to 1 core each.
We're missing an important piece of the picture here, which is ... I have no idea what CPU you have. But if you have a 4-core hyperthreaded CPU (e.g. a 4-core i7) and you're making this CPU assignment strategy, then yes this is a dumb way to go about it -- 2 of the game instances would be restricted to using a logical ("fake") CPU core.
Most people just pick one of the options in the CPU Strategy wizard (or the page in the Quick Setup Wizard). The CPU Strategy Wizard has a Hyper-Threading option, specifically for CPUs that have logical cores, which pairs physical and logical cores together for improved performance.
Here's an improved version of your CPU strategy:
Char 1: Core 1,2,3,4
Char 2: Core 5,6,7,8
Char 3: Core 5,6,7,8
Char 4: Core 5,6,7,8
Char 5: Core 5,6,7,8
